On theoretical grounds, overdosage would be expected to cause enhancement of the pharmacological side effects mentioned previously. Observations in animals show that extreme overdosage (100-200 times recommended daily dose) may produce oestrogenic effects. There have been reports in the literature that Tamoxifen (NOLVADEX-D) given at several times the standard dose may be associated with prolongation of the circulating level of estrogen from the pituitary gland ( vaccines) and with consequent and onset of ovarian enlargement. In order to extend the duration of treatment, several weeks (perhaps up to two years) after treatment have been stopped, a single daily dose of tamoxifen should be taken to promote maximal effective dose of estrogens.
